StrataScript® one-tube RT-PCR system offers high efficiency TA/UA cloning with high fidelity in a one-tube RT-PCR format. The simple format includes our robust StrataScript® reverse transcriptase for 1st strand cDNA synthesis and Easy-A™ PCR cloning enzyme, which has equivalent fidelity to Pfu and PfuTurbo® DNA polymerases, for amplification.


Robust High Fidelity Amplification in Single Tube RT-PCR

The new StrataScript® one-tube RT-PCR system is ideal for fast and easy one-tube set-up for RT-PCR. The simple one-tube format makes preparing multiple reactions fast and easy and eliminates carryover contamination. The kit features our robust StrataScript reverse transcriptase for 1st strand cDNA synthesis. StrataScript RT is a nuclease free enzyme, which delivers robust yield and all around performance needed for RT-PCR. The one-tube system also features our Easy-A™ PCR cloning enzyme, which is 6-fold more accurate than Taq DNA polymerase and can amplify a wide variety of target lengths from 0-5 kb.

Same Cloning Efficiencies as Taq

Products amplified with Easy-A PCR cloning enzyme are cloned into T- or U-vectors as efficiently as those amplified with Taq DNA polymerase. Table 1 compares the cloning efficiencies obtained for 600-bp to 3-kb fragments amplified with the Easy-A high-fidelity PCR cloning enzyme or Taq DNA polymerases from two commercially available sources. The data shows that using Easy-A enzyme in a one step RT-PCR format delivers the same cloning efficiencies as using Taq DNA polymerase.
